Cellulose is the most abundant organic raw material on the Earth, and it is the most renewable resource that is available in large quantities. Presently, only a few cellulose are utilized effectively, most of them are wasted and pollute the environment.In this paper, five microorganisms producing thermophilic cellulase was screened from manure of cattle, compost and soil. One of them, which can grow better and produce more cellulase on CMC plate is selected for further research, and named GPL1. GPL1 is a gram positive aerobic actinomycete. The GPL1 was identified according to the physiological and biochemical properties, and the 16S rDNA sequence and systematic analysis. It was most similar to Thermobifida fusca YX, and was initially defined as Thermobifida fusca GPL1.The cellulase produced by Thermobifida fusca GPL1 consisted of Cx, C1 and β-glucosidase. In order to improve the cellulase production of Thermobifida fusca GPL1, the medium components and fermentation conditions of the three enzymes were optimized. After optimization, the optimal mediums is:Sugarcane bagasse powder 1.3% Yeast extract 3.0% NaCl 0.8% MgS04 0.05% KH2P040.6% CaCl2 0.01% pH 7.5.The optimal fermentation conditions is:Liquid volume 60ml/250ml (or 120 ml/500ml), Inoculation 12%, 65℃, 200r/min, 50 h;The enzyme activity after optimization are that: Cx, 10.1 IU/ml; C1, 0.056...
