Microemulsion is prepared with appropriate water, oil, surfactant and cosurfactant and with special technics, whose particle size is between 10 and 100nm, whose appearance is transparent, whose different direction is parallel and whose thermodynamics is stability. As one kind of drug carrier, microemulsion has many advantages such as low viscosity, good stability, increasing incorporation of drugs that do not dissolve in water, improving stability and scatter of drugs, accelerating absorbability of organism to drugs, enhancing biologic utilization of drugs. The objective of this experiment is to prepare oil-in-water rifampicinum microemulsion and investigate rifampicinum icroemulsion's stimulation to animal's skin.In the experiment surfactants of Tween-80, Span-80, Glycerol monostearate, Simpsoltmmsf3020, Emulsifier340B, cosurfactants of glycerine, alcohol, synth eticnbutyl alcohol, propanediol, 1,3-butyleneglycol, non-ionic water and some oil of Mineral oil, Isopropyl myristate, Soybean oil, Jojoba oil, VitaminE, Dimethyl silicon oil were chose to prepare microemulsion by emulsification of phases transforming. On the base of microemulsion recipes, pseudo-ternary phase diagrams were depicted with titration and the distribution of microemulsion was determined. Sieve the best recipe and investigate it's incorporation to rifampicinum. By emulsification of phases transforming, rifampicinum microemulsion was prepared. Using ultraviolet spectrophotometry, drug consistences of rifampicinum microemulsion were mensurated; Using transmission electron microscope, particle shape and size of microemulsion and rifampicinum microemulsion were determined; Investigate stability of microemulsion and rifampicinum microemulsion by accelerated testing in constant temperature and reserved testing in different temperature; Investigate stimulation of microemulsion and rifampicinum microemulsion to animal's skin by stimulated testing to rabbits'skin.Outcome show that when the ratio of Tween-80 and Span-80 were 5:1, 4:1, 3:1, 2:1,these surfactant can be prepared to microemulsion with Mineral oil, Isopropyl myristate, Mineral oil/Jojoba oil/VitaminE by emulsification of phases transforming. From pseudo-ternary phase diagrams that had been depicted, that microemulsion area is biggest when the oil is Mineral oil can be seen.
