In this paper,the preparation of tilapia skin collagen enzymic hydrolysates,the antioxidant properties in vitro of tilapia skin collagen hydrolysates,and the purification of superoxide radical scavenging active peptides from tilapia skin collagen hydrolysates were investigated.(1)The tilapia skin was dealed with acid solution and alkali solution. then rinsed with distilled water at 45℃for 12h.The supernatant was lyophilized and referred as the tilapia skin collagen.The yield of co]lagen was of 13.6%(on the wet basis),the purity was 89.87%and the protein content WaS 95.05%(on the dry basiS).Bromelain,Alcalase, trypsin,neutral protei nase,pepsin,flavor proteinase and alkali proteinase were used to hydrolyzed the tilapia ski n collagen.The results showed that the hydro]ysates by bromelain and the hydrol ysates by Alcalase exhibited higher superoxide radical scavenging activity than the others. So the optimal conditions of the hydrolytic reaction of bromelain and Alcalase were detez'mined by the orthogonal test L16(45).The results showed:for bromelain,45℃,pit4.5,2000U/g(E/s),4h, 6%(substract concentration);for Alcalase,55℃,pH7.5,3600U/mL(E/S),2h,4%(substract concentration).In order to obtain enzymic hydrolysates with high superoxide radical scavenging activity,compound enzymic hydrolysis was adopted on the basis of single enzymic hydrolysis.The optimal compound hydrolytic technology was first hydrolyzed by bromelain then by Alcalase, with their optimal hydrolytic conditions.The superoxide radical scavenging activity of the enzymic hydrolysates by optimal compound hydrolytic technology was very high,its inhibitory value was as high as...
