Gymnema sylvestre, a plant native to the tropical forests of south of China, has Gymnemic acid can used to prevent and treat diabetes.Gymnemic acid can be determined by spectrophotometry of dealt with mixed liquid of 8% vanilin-sulphuric acid.The output of gymnemic acid was regarded as an examine target and the influences of soaking time, soaking temperature, ratio of material to water and extractable solvent on the extraction have been studied. The orthogonal test were used to determination the best extraction process was as follows: the leaves of Gymnema sylvestre were extracted with 50%ethanol, 70℃, the ratio of material to water is 1:15 for 2h.This paper was studied on used resin to improve the content of Gymnemic acid after determination the best extraction process. The result of saturated static adsorption of different types of resins showed that YWD-04 was suited to use on Gymnemic acid. Through the studied of dynamic adsorption, YWD-04 had the best adsorbilities when the concentration of Gymnemic acid was 1 —2.5mg/mL, the adsorption current velocity was 1 — 2BV/h. Meanwhile, through the studied of dynamic desorption, YWD-04 had the best desorbilities when the volume of 60— 80% ethanol as eluant with desorption current velocity was 1 — 2BV/h. Gymnemic acid was greatly refined by resin.The animal test results showed that Gymnemic acid could significantly decreased blood glucose level of the diabetic mice induced by alloxan by Gymnemic acid 100 mg/kg weight everyday. The orthogonal test were used to determination the best prescription for diabetes mellitus with Gymnemic acid.
