5'-Adenosine triphosphate is used to synthesizephosphatidate(phosphatidylcholine, phosphatidylserine, phosphatidylserine, phosphatidylethanolamine, etc). It was widely used in medical industry and chemical industry, especially applied in the field of medicine.Presently, few civil companies could produce 5'-Adenosine triphosphate because of low purity and yield. Consequently, it can't meet the demand of medical industry. Therefore, we must import about 50 tons of 5'-adenosine triphosphate every year. The shortage of 5'-adenosine triphosphate has restricted the use and development of it in medicine and fine chemical fields.Based on the viewpoint of industrialization of 5'-Adenosine triphosphate, this paper was studied the separation process of 5'-Adenosine triphosphate by using of cheap and effective ezymohydrolysis-ion exchange separation.Because the physical and chemical properties of 5'-Adenosine triphosphate, 5'-Adenosine disphosphate and 5'-Adenosine monphosphate are very similar, the separation is greatly difficult to carry out. At civil, the shortcoming in separation is that the resolution and using ratio of resin is low, the amount of pigment contained in products is too high, and the purity and yield of 5'-Adenosine triphosphate is too low.Aiming at above problems, the strategy of separation was put forward as follows. Firstly, in view of great amount of protein and impurities in solution, the quality of product is declined. So we deposit the protein by hydrochloric acid, which makes 80% protein be removed. Secondly, on the basis of physical and chemical properties of AMP, ADP and ATP, we first designed and synthesized several kinds of anion exchange resins, and screened out an anion resin-SD3 which possessed high adsorption capacity, high separation factor for 5'-adenosine triphosphate, and high removal ratio of impurities.
