5'-Cytidine triphosphate is used to synthesize phosphatidate(phosphatidylcholine,phosphatidylserine, phosphatidylserine, phosphatidylethanolamine and so on). It haswidely been used in the industry of medicine and fine chemicals, especially applyingin the field of medicine, which are unreplaced.Presently companies inland that produce of 5'-Cytidine triphosphate are very few,and the purity of products is not high, and yield is low. Every year we must import alot of 5'-Cytidine triphosphate, which can't meet the demand. The shortage of stuffhas restricted the use and development of 5'-Cytidine triphosphate in medicine andfine chemicals.From the viewpoint of industrialization of 5'-Cytidine triphosphate, this articlestudied the production method of 5'-Cytidine triphosphate taking advantage of cheapzymohydrolysis-ion exchange separation.Because the physical and chemical properties of 5'-Cytidine triphosphate,5'-Cytidine disphosphate and 5'-Cytidine monphosphate are very similar, and thereare many impurities in the solution of zymolysis, the separation of down stream isgreatly difficult. At present inland the shortcoming in separation is that the resolutionand use ratio of resin is low, the amount of coloring matter contained in products istoo high, and the purity and yield of 5'-Cytidine triphosphate is too low and etc.Aiming at above problem, this paper put forward the strategy of separation asfollows. Firstly, in view of great amount of protein and impurities in solution, thequality of product is declined. So we deposit the protein by hydrochloric acid, whichmakes 80% protein matter be removed, and then we Secondly, on the basis of physicaland chemical properties of CMP, CDP and CTP, we first synthesized several kinds ofanion exchange resins by ourselves which are used to separate them preliminarily. Wesynthesized several differently qualitative anion resin, and screened out an anionresin-SD3 which possesses high adsorption capacity, high separation factor for5'-Cytidine triphosphate, and high wiping off ratio of impurity. The adsorptioncapacity of resin SD3 to 5'-Cytidine triphosphate is 0.2075g/g.wet resin, the iABSTRACTseparation factor ?CTP is more than 5.0, the wiping off ratio of impurity more than 后杂45%. The separabilities of resin SD3 toward CMP, CDP and CTP are high. Theamount of impurity in the solution of CTP is great, which is removed difficultly. andthe adsorption isotherm line f"(c)<0 is favorable for adsorption equilibrium. From thekinetic and thermodynamics behavior of resin SD3 adsorbing 5'-Cytidine triphosphate.We learned that when the concentrations of 5'-Cytidine triphosphate are 1g/L and7.5g/L, the diffusions come under FDC and PDC respectively. At the same time, weresearched roundly the dynamic process of resin SD3 adsorbing, eluting 5'-Cytidinetriphosphate, from which we learned several technological conditions of resin SD3purifying 5'-Cytidine triphosphate, and we get 5'-Cytidine triphosphate with thepurity 96 % and the yield 95.5%. All of these haven't been reported. Finally wepreliminarily researched the simulation of the adsorption process of 5'-Cytidinetriphosphate in different conditions.This method possesses that procedure is simple, the demand for equipment is low,and the cost of production low, and is propitious to the demand of wholesaleindustrialization.
