Since the technology of immobilization was invented, it has been applied into many fields, such as food, chemical industry, medicine, chemical analysis, environment protect and the exploitation of energy. But, there is a big distance between home and abroad. In foreign, the technology of immobilization has been applied in the industrialized production, and the using cycles of some immobilized enzymes or cells were much longer than that in China. It is much later to research technology of immobilization in China than in foreign and few factories or plants use immobilization to biocatalysis and biotransformation, for that the reclaim rate of enzymatic activity is low in single-immobilization and the reaction ability is bad. So, there is far-reaching realistic meaning for feather researching the technology of immobilization.Integrated immobilization is researched that a system of coupling-immobilization is formed by integrating other immobilizing methods or adding some substances on the basic of traditional immobilizing methods, to conform a micro-environment for bioactive substance in existence stably. The micro-environment could avoid the effect of outside environment to biocatalyst and maintain enzyme intrinsic structure and functions and exert all enzyme catalysis ability long and overcome the disadvantages of single-immobilization. The immobilized enzymes or cells would be gained with high stability and high bioconversion rate of enzyme activity, to apply into industrialized production.First, the immobilized gel of carrageenan was optimized by adding different additive or enzymatic protecting substance in the mix-gel of carrageenan and gelatin, to reduce the carrageenan gel's freezing point and to protect the enzyme activity. The coupling-immobilization system having 3.5:0.5 of carrageenan and gelatin, 0.05mol/L trehalose and 2×10-3mol/L Mg2+ was applied for L-Phe production.
