| The volatile ingredient in traditional Chinese drug is widely applied to preparations owing to its effects of antibacterial, antiinflammation, acesodyne, etc. During the production and the storage, they degrade very quickly, which leads to diminish the curative effect and affects the appearance of the preparations. So it is necessary to study on the stability of the volatile ingredient in the preparations. In this work, the main goal is to prepare the BS microcapsule, which can not only improve the BS's stability and fluidity but also step up the delivery and absorption of BS. The microencapsulation provides the method and the way of thought for resolving the problem of the volatile ingredient in traditional Chinese drug. This work consisted of four production as a whole.First, a solvent-nonsolvent method was employed to prepare the BS microcapsules. The influential factors of preparation were studied. The preparation method was optimized according to an orthogonal design of experiments. The optimum preparation method was: the ratio of BS dispersion: PEG-6000 is 1:4, concentration of SPAN80 is 1.5% and temperature is 50℃, which was stabile and dependable.Secondly, investigation and evaluation of BS microcapsule was carried out on the aspects of powder technology, stability, dissolution. The tests of powder technology showed that microencapsulation improved greatly the fluidity and performance of fill of BS and made the process of pharmaceutical preparation easier. Dynamics parameter of the classic test of constant temperature indicated that microencapsulation increased greatly the stability of BS. Experiment on the release in vitro showed that the release rate and bioavailability of BS was increased greatly by the microencapsulation.Thirdly, the assay of the content and dissolution of BS in the microcapsule was separately established. The results of verification test showed GC with a capillary column which measured the content and dissolution of BS in the microcapsule was reliable and accurate. The internal standard was menthol. The method of solvent extraction was simple and complete, which may server as the extraction and... |
