Ginseng is a traditional medicinal material in our country, the ginsenosides are the main active components of ginseng, ginsenoside Rb1 has extensive pharmacological effect such as reducing cholesterol density, strengthening memory, anti-oxidation, etc. Meanwhile, ginsenoside Rb1is raw material of the anticancer drugs Rg3 and Rh2, and it plays an important role in ginsenosides.Now, the macroporous adsorption resin method and the traditional extraction by methanol or ethanol are the primary method for separating ginsenoside Rb1, which could not be applied in a large scale and so could not satisfy the practical need.In this paper, the separation of ginsenoside Rb1 coring by Simulated Moving Bed Chromatography(SMBC) and the possibility of separating ginsenoside Rb1 by SMBC were studied. The whole process of separating ginsenoside Rb1 with SMBC was discussed from the theoretical and experimental sides. Based on the experiments, the pre-treatment conditions of ginseng raw material and the post-treatment of SMBC products were optimized. Notoginsenoside, ginsenoside raw materials from FuSong and CAAS (China Agricultural Academy of Science), their SMBC separation were compared. The works are as follows:Part 1: Pre-treatment conditions of ginsenoside raw materials were optimizated, the separating parameters of SMBC were selected and the extraction conditions of SMBC product in the post-treatment were selected. under the optimized conditions, 92% purity of ginsenoside Rb1 can be obtained. The working parameters are: mobile phase: V(methanol):V(water)=45:55; Dilution rate: UD = 60 mL/min , Raffinate rate:32 mL/min ; Extract rate: 31wL/min ; Injection rate: 0.2g/mL ; Injection rate: 3 mL/min; Switch time: 6 min; column configuration: 1 - 4 - 3.Part 2: Three different kinds of ginsenoside raw materials were applied in the experiments and the results showed the good results for the ginsenoside raw materials from CAAS and notoginsenoside raw materials were obtained by SMBC.Part 3: The SMBC product was identificated by UV, IR, MS and NMR. 92% purity of ginsenoside Rb1 was obtained.Part 4: The techniques of separating ginsenoside Rb1 with SMBC and single column were compared.
