To further study total fatty soluble biological activity seabuckthorn composition, seabuckthorn fruit was made into seabuckthorn total fruit oil by supercritical CO2 extraction technology and get high quality total fatty soluble activity seabuckthorn composition for one time. Also the function of immunoregulation and liver protection were studied in this paper.First of all, the contents of isorhamnetin of seabuckthorn fruit from different producing area were determined by HPLC. Isorhamnetin and quercetin were determined by TLC. And the standard was used to control the quality of seabuckthorn fruit. Second, supercritical CO2 extraction technology was applied to extract seabuckthorn total fruit oil. The optimum extraction technique parameters were as follows, rate of flow was 20L/h, extraction pressure was 30Mpa, extraction temperature was 45℃, extraction time was as long as 3 hours. The extraction rate was above 18%. In production, a heavier outflow was adopted to shorten extraction time.The maximum toleration volume of seabuckthorn total fruit oil was above 20000mg/kg.bw to the mice and rats of different sex, the acute toxicity and heredity toxicity experiments proved that the three mutagenesis experiment results were negative and the oil was innocuous.The immunoregulation function experiments such as the test of half hemolysis showed seabuckthorn total fruit oil could thicken the soles of the rats, improved the reproduction ability of the lymphoid cells, increased the cells produced by spleen and half hemolysis, and improved the phagocytic function of the macrophage, after the oil was given to the rats for 30 days. So it could be deduced that seabuckthorn was of immunity function.The liver protection experiment was the test of ALT and AST of rats'serum and so on. The rats were divided into two groups in the liver protection experiment. The two groups were given CCl4, group one was given varying amount of the oil. The liver damages to group one which was given varying amount of the oil were less than group two. After the experiment data was processed by statistics, the results of the two groups were of great difference. This was because the oil played the better part of protection the rats'liver than seabuckthorn seed oil.
