Preparation Process Of Exopolysaccharide And Cordycepin From Solid Culture Medium Of Cordyceps Militarism Link

Solid medium method has been implemented widely in the culture field of Cordyceps Militarism Link in recent years, there are large amount of myceliums grow in the solid culture medium in cultivating process, but for there no systemic process to extract the active components of the culture medium, most of them are being rejected for waste, it lead to the waste of resource, and threat the environment synchronously, by the development of Cordyceps Militarism Link cultivating domain, it's became urgently to process the solid culture medium of Cordyceps Militarism Link.Focus on developing solid culture medium of Cordyceps Militarism Link and different physical and chemical characters of the active components, process conditions which influencing the extraction and separation were studied. Orthogonal design was be used to assess the influence of temperature, times, cycle time and ratio of the raw material and water, final optimized conditions for extracting exopolysaccharide was established, extracted 3 times by 15 times of water, and extract 120 mins each time on 80oC further Sevage method, Developed Sevage method and Clarifier method were used to separate the protein, DEAE cellulose chromatograph and polysaccharid gelatin chromatograph were experimented to purify exopolysaccharide. Focus on research the optimized process conditions to extract cordycepin, orthogonal design was also be used to assess the influence of the concentration ethanol, extracting times, cycle time and the ratio of the raw material and solvent, finally excellent extraction process condition of cordycepin was established, using ultrasonic to extract 3 times in 20 times of 75% ethanol solution, and last 40 mins each time, further ion exchange chromatograph and silica gel chromatograph were used to purify cordycepin. Purification analysis on exopolysaccharide and cordycepin had also been launched respectively, phenol-sulfuric acid method and HPLC were used to qualified the difference of several purifying processs.Based on the results of the lab researches, focus on the synthetically extraction of exopolysaccharide and cordycepin from solid culture medium of Cordyceps Militarism Link ,three enlarged batch experiments were valided to extract exopolysaccharide and cordycepin, the process condition and route were established , the route is, circumfluence pre-extraction by water, deposit exopolysaccharide by ethanol and separate protein by sevage method , extraction and purify cordycepin by aether and negative ion exchange chromatograph. The purification of exopolysaccharide(protein separated) are beyond 50% and content of purified cordycepin were beyond 97%.This study comes from the practice of the culture field for Cordyceps Militarism Link, the results would be provided as technical support for relative enterprises.
