Glucosinolates are abundant plant secondary metabolites that are found in cruciferous plants. They have much effectiveness as a pesticide, anti-microbial reagent and natural fumigated disinfector. Isothiocyanates are their degradation products, which have shown a series of biological and chemical characteristics. Besides their special flavor for condiments, they have many biological properties such as anti-bacterial, anti-carcinogenic potential. ITCs can protect DNA-damage and cancer caused by many dietary carcinogens such as polycyclic aromatic hydrocarbons, heterocyclic amines and nitrosamines. These properties have attracted the interesting of many food chemists, medicinal scientists and biologists.In the present article, we give studies on the formation, chemical properties and discuss extraction conditions. The optimal conditions were that proportion of water and horseradish, temperature and time of extraction were 70 mL/g, 90℃and 20 min respectively. The optimal condition of detection were that temperature and time were27℃-29℃and 4h. By comparison studies on 12 macroporous resins adsorbing or desorbed the glucosinolates in horseradish, D201GF that is the most strongly basic anion exchanger-resin among 12 resins, shows the higher adsorption capacity and easier desorption to glucosinolates than others. Then and it can be used to extract the glucosinolates from aqueous solution. Effects of the volume of the extraction solution, pH value, temperature and time of adsorption on the recovery of glucosinolates were studied. Meanwhile, the influences of varieties of elution, the minimum amount and time of desorption on the desorption method were also evaluated. The results show that maximum adsorption of D201GF is 23.94μmol/g and the adsorption yield is 96.26%. The effect of pH value was no significant. Therefore, pH-adjust is not necessary. The optimal conditions were that temperature and time of adsorption were 50℃and 15 min respectively. The optimal eluting solvent was 1 mol/L of sodium chloride. The desorption yield is excess 99% in the addition of temperature of 50℃and shaking time of 15 min. The extracting solution of glucosinolates was purified, a reliable method to separate and identify glucosinolates in horseradish was developed by using reversed-phase ion-pair HPLC. With the shooting of horseradishand, Effects of temperature, time, pH value, concentration of Vitamin C on hydrolyzation of glucosinolates were studied. Designing orthogonal experimentation, the optimal conditions were that temperature, time, pH value and concentration of Vitamin C were 30℃, 20 min, 4.0 and 0.2 mg/g respectively.
