With the improvement of living standard and changing of diet, the incidence of diabetes mellitus has got higher and higher, they are becoming serious danger for humankind. Chinese traditional medicines have tremendous potential on curing diabetes mellitus. From the 1980s, experts all over the world pay attention to mulberry leave for its remarkable effect on curing diabetes mellitus. Dwarf lilyturf tuber is also one kind of Chinese traditional medicine, having much medicine action, especially the effect of curing diabetes mellitus, which is always the hotspot of study. Though much research has been done on the pharmacologic actions of mulberry leaves and dwarf lilyturf tuber in reducing blood sugar, few have been done on the extraction technology in this regard.In light of this, we concducted experimental research on the extraction methods of ophiopogonins, DNJ, polysaccharide in the paper by using such advanced techniques as microwave-assisted extraction, enzymatic method, aiming to find an optimal extraction technique. In addition, we compared the reflux method, enzymolysis method and microwave-assisted extraction method in the study. The experiment results were analyzed statistically and intuitively to find an extracting method that is significantly better than the normal method in the optimal condition, thus, greatly reduing the extraction time. The major contents and results are as follows:1. Using different solvents to extract ophiopogonins and DNJ. Of them, enthanol has a better extraction effect than water. Given to the fact that there is no significant difference in the extraction rates between using enthanol and water, and for the sake of cost, the present experiment adopts water extraction.2. Comparing several extraction methods and optimizing the experiment. Several methods such as refulx, microwave-assisted extraction and enzymatic method are used to extract ophiopogonin, polysaccharide; and also, single-factor experiment and orthogonal methods were used. The optimum extraction condition in microwave-assisted extraction is: 10min, 36% power, the ratio of solid to liquid is 1:14. The extraction rate of ophiopogonins and polysaccharide under this condition is 2.323mg/g and 8.875mg/g respectively. This is a great reduction of time and an increment of extraction rate compared with the traditional reflux extraction and enzymolysis method (120min, 45°C, enzyme, extraction rate: 2.265mg/g and 9.854mg/g respectively).3. Comparing several extraction methods of DNJ, polysaccharide and optimizing of experiment. Microwave-assisted extraction, enzymolysis method and reflux method were used to extract DNJ and polysaccharide, and orthogonal methods were used compare the difference between these methods. The microwave-assisted extraction and enzymolysis method were better than the traditional reflux method concerning their extraction rate. microwave-assisted extraction was more advantageous in terms of time consuming, energy consumption and extraction rate, as the extraction rate of DNJ and polysaccharide are 1.102mg/g and 4.755mg/g respectively. Yet, microwave-assisted extraction was not so remarkable in the extraction of ophiopogonins and polysaccharide, compared with other methods (enzymatic method and reflux method).Applying the microwave-assisted extraction to the Chinese herb (especially the trace and residual component) can significantly reduce the extraction time, save dissolvent, yet obtaining the ideal extractions with higher rates.
