Salvia miltiorrhiza is one of the most important tradition Chinese medicinal materials that possess the function of both promoting blood flow and removing blood stasis with favorable anti-coronary artery disease. The consumed quantity only in the aspect of injection has been exceeded 2 billion dosages every year. However, because the undetermined active ingredients and relaxed index of quality control induced the instable effects of clinic traterment, and, moreover, usually adverse functions, which lead salvia miltiorrhiza unable to accommodate requirement of contemporary Chinese medicinal materials melting world.Sulfotanshinone Sodium is a sodium chloride of water-solubility after sulfonating tanshinone IIA of liposolubility active ingredient abstracted in salvia miltiorrhiza. It is an effective medicine of angiocardiopathy to treat coronary artery disease, angina cordis, and myocardial infarction. The amount of clinical utilization continuous increasing with the high proportion of patients of angiocardiopathy induces raw material resource of salvia miltiorrhiza insufficient in China. So it becomes an extensive focus point in pharmaceutical factory that sufficient utilize and cut down utmost dissipation resource of salvia miltiorrhiza. The major idea of this work has investigated the influencing factors of the amount of glacial acetic acid and acetic anhydride, the reaction temperature, the quantity of mixed acid, the ratios of mixed acid, the droping speed of mixed acid, and the time of agitation and so on the yield of Sulfotanshinone Sodium in the synthesis process, based on systemicly surveying its pharmacologic action, pharmacokinetic and preparation technology. Thereafter, the orthogonal optimization experiment is conducted according to7 factor 3 level layout [L18(37)]. The obtained optimal synthesis conditions are as follows: Given 0.5 g of tanshinone IIA, the amounts of glacial acetic acid and are 2.4mL and 4.0mLrespectively; The reaction temperature is 10°C; The quantity of mixed acid is 2.0mL; The ratios of mixed acid is CH3COOH: H2SO4 = 1:2; The droping speed of mixed acid should be controlled at 2.5 mL/min; and the agitation should be kept to 1 h. The yield of Sulfotanshinone Sodium prepared under this conditions reach up to 66.3%.
