In this paper, two novel oil soluble chitosan derivatives—OACS and OAGCS were prepared by introducing oleic acid onto 6- hydroxyl groups of chitosan and EACS, respectively. The amino groups were protected locationally. The structure, crystallability and thermal property were characterized by FT-IR, 13C NMR,1H NMR and XRD. The solubility in general organic solvents, oleic acid and lipin of both derivatives were also studied. In order to find out the solubilities in general organic solvents, PhCS that had satisfied solubility in DMF, DMAC and DMSO and EHCS that was amphiphilic were prepared. The derivatives were selected as probes to study the absorbability and rheological property in free fatty acid, unsaturated fatty acid and heavy metal ion. Also in this paper, there is a preliminary research of absorbability in lipin, lipoprotein and biliary acid of the derivatives.The results show that the preparations mainly took place on 6- hydroxyl groups of chitosan because of the protective step. The solubility of OACS and OAGCS in oleic acid and lipin is much better than that of raw chitosan, and OAGCS is the best one. The chelated efficiency of derivative OACS, OAGCS and EHCS with Cu2+ and Pb2+ has increased and EHCS has an absorbability as much as twice of that of chitosan. Compared to chitosan, OACS and OAGCS have apparently enhanced the absorbability in lipin. The derivatives can be added into edible oil without disrupting whose rheological behavior, meanwhile they can absorb lipin and partly unsaturated fatty acid. The absorbability of chitosan and its derivatives has the order of OACS, OAGCS, CS and EHCS. However, OACS and OAGCS have lower absorbability in diversified lipoprotein. EHCS has an increased one compared to raw chitosan but a poor selectivity with HDL.
