In this paper, two novel oil soluble chitosan derivatives—ONNCS and DMOACS were prepared through introducing oleic acid and N,N dimethyloctadecyla quaternary ammonium salt group onto the 6-hydroxyl of chitosan respectively, with the locational protection of amino group. Because of the presence of some harmful substances, such as free fatty acids, some kind of unsaturated fatty acids, heavy-metal ion, and aspergillus flavus, the absorbability and rheological behavior of ONNCS in the edible oil were tested. Moreover, the absorbability to lipoprotein and bile acid of DMOACS were also studied. Finally, the two oil soluble derivatives were researched in animal experiment with the treatment and precaution methods respectively. In the result, ONNCS can solve in oleic acid with a solubility of 3g, and swell in lard oil and edible oil. DMOACS has a good solubility in DMSO and deionized water. However, the destroy of crystal structure also decreases their thermodynamical characteristic. ONNCS can selectively absorb unsaturated fatty acids and lipid in edible oil with the reservation of oleic acid which is beneficial to health. What's more, ONNCS also has an excellent chelation to pb2+ and Cu2+. ONNCS has little effect to the rheological behavior of oleic acid with a concentration below 1.5%, however, when the concentration is below 1.5%, it will improve the fluidity at 300C. The absorptivity to LDL of DMOACS is not improved conspicuously, but the one to HDL is higher than 85%. Comparing with chitosan, DMOACS has a higher absorbability to bile acid, but the one of ONNCS is lower. And the absorptivity is in direct proportion to the initial concentration. Both of the two methods of animal experiment show that the two oil soluble chitosan derivatives can significantly lower the body weight and body fat content. They can lower the serum lipid, glucose and LDL of rats and increase HDL to different level while increase the feces fat excretion at the same time. Because all of these characteristics, with the nontoxic and good biocompatibility of chitosan, DMOACS is expected to be used in the treatment and precaution of obesity, high blood fat, atherosclerosis, and their syndromes with the absorption of bile acid and serum lipids. ONNCS can be used to adjust the content of edible oil and clean the heavy metal ions and lipids in the body and edible oil.
