LtHb is extracted and purified from earthworm lumbricus body through the methods of aqueous two-phase and Q-sepharose fast flow ion exchange chromatography. The studies on the condition of purification from earthworm lumbricus terrestris body and the character of its and on the modification of LtHb using TPP and ACS that have been synthesized by using acrylic acid and chitosan.The studies have been done on the preparation of LtHb nanoparticles.The experimental results indicate that the condition of purification from L.T. depends on the isoelectric point and the pH of sulfate system and so on.The products of LtHb is very pure and the function well as oxygen carrier through the kinds of experiments ,for instance,SDS-PAGE,the content of heme and the animal expriment.N-carboxyethylchitosan have showed that excellent biogradable properties with standard activated sludge.In the paper, the reaction conditions of N-carboxyethylchitosan have been systematically investigated through orthogonal design,the best conditions have been found.The experimental results have showed that the average substitution degree of amid of chitosan is mostly affected by the ratio of acrylic acid and chitosan,the reaction temperature and the reastion time. The products have been characterized by ninhydrin assay ,FTIR and ~1HNMR.The nanoparticle of LtHb have first prepared by using ACS and TPP in the study.The production have been obeserved using TEM.The experiments have been done to study the LtHb-encapsulation ratio and the LtHb-carried amount ,the LtHb release behavior in the sulfate and simulation gastric juice have been done too.
