Experimental Study On Soybean Isoflavone Extraction

The extraction procedure of an active substance in soybean -- isoflavone by solvent extraction was studied in the present thesis. The extraction experiment via supercritical carbon dioxide was elementarily discussed as well.The method of analyzing the content of soybean isoflavone by High-Performance Liquid Chromatography (HPLC) had been established with American Waters 1525 series HPLC. Chromatography conditions were asμBondapah C18 reversed-phase chromatographic coloumn (10μm particle size, 300mm×4.6mm). Injection amounts of standard sample were 5, 10, 15, 20, 25, 30μL, while experimental sample was 10μL. Mobile phase was Methanol: 0.3% Phosphoric acid =1:1.Flowrate was 0.7ml/min.Column temperature was room temperature. Analyzed time was 45min and detection wavelength was 254 nm. With selecting defatted soybean cake as raw material and ethanol as solvent, single element effect test was conducted under four factors, namely, content of ethanol, soaking temperature, soaking time and liquid-to-solid ratio. At the same time, the influence of these factors to the amount of solute was analyzed. Next, the orthogonal test was conducted and the optimal condition was found as soaking temperature 70℃, liquid-to-solid ratio 1: 20, soaking time 6h, content of ethanol 80% and the extraction rate 0.2%. As one part of the thesis, the kinetic equation of isoflavone solvent extraction was validated by analyzing others'data, and according to the test results in the present work, and the kinetic model equation was correlated. In this part, the phenomenon of isoflavone decomposition as time going on was discussed consequently.Supercritical carbon dioxide extraction for isolating the isoflavone was conducted at the pressure varied from 30MPa to 60MPa. It was found that with the pressure increasing, the solute increased, but the extraction rate was lower than that of solvent extraction.
