The hydrogen peroxidase is also called catalase(CAT). As one kind of oxidases, catalases can be found in almost all animals, plants and microorganisms. It decomposes hydrogen specifically. It dissolves to water, nearly not dissolve to ethanol, chloroform, and ether. Catalases can catalyse the decomposition of the cellular hydrogen-peroxide and thus protect membrane lipid against peroxidation. The research of catalases can be run down until the beginning of 19 century. Thus far, catalases is used in agriculture, food, paper factory and so on.Firstly, catalase from pig blood has been purified by the use of sephadex G-100 and PEG-6000 chromatography. The result is that 0.64 g catalase can be purified from 100 mL pig blood cell. The total activity is 109037 mmol/min/mL, the activity of rough catalase is 29796 mmol/min/mL, and the activity of rarefied catalase is 87448 mmol/min/mL. The callback rate is about 80.2%.Secondly, through the single-factor experiments and orthodox experiments, this paper studied on the method of testing the catalase's activity. The wavelength is 405 nm, the pH is 7.4, and the time of the reaction is 3 min.Finally, the characters were studied. This paper studied on the influence of temperature, pH, NaN3 and KCN, and different metal hydroniums. Through SDS-PAGE experiment, sub-radix molecular weight was tested. Saecharide of catalase was tested by phenol-vitriol method.
