Thymopentin, also known as TP-5, is a synthesized derivative of thymopoietin, a naturally occurring hormone responsible for inducing T-cell precursors to differentiate and mature. Thymopentin (TP-5) is a short-chain peptide which is composed of five amino acids. Thymopentin is mostly used in curing cancer, herpes simplex, rheumatoid arthritis, AIDS, severity immunodeficiency syndrome, chronic hepatitis II on clinic.In this paper, the synthesis and separation methods of thymopentin were studied. The methods were optimized and scaled up. The main content is following:1. Firstly, general methods for SPPS, current TP5 synthesis level and its disadvantages had been reviewed. Secondly, several following key steps involved in SPPS were particularly studied: swelling of resin; analyzing the coupling time; mole ratio of input Fmoc-amino acid; selecting the coupling reagents in the elongation; cleavaging peptide-resin. The high-efficient and low-cost TP5 synthesis process had been established, which indicated perfect feasibilities to the large-scale synthesis.2. The separation and transformation conditions of thymopentin were studied on ion-exchange chromatography and reversed phase chromatography was applied to prepare refined TP-5. After freeze-dried, TP-5 was analyzed with HPLC, MS, IR and NMR. Finally, we scaled up the method, the purity of TP-5 is more than 98% and the total yield is 63%.Eventually, there had been established one high-efficient and low-cost TP5 synthesis, purification, transformation and refination process. The proposed optimum procedures had showed their feasibilities and benefits in both small and large scale TP5 synthesis process.
