Cephalosporin C is a kind ofβ-lactam antibiotic produced by some filamentous fungus such as Cephalosporium acremonium. 7-Amino Cephalosporanic Acid, the degraded product of cephalosporin C, is one of the important intermediates ofβ-lactam antibiotic.In this paper, a strain screening model of metabolic control was set up and put up to application with Cephalosporium acremonium MNS-A8-51 as the original strain. In addition, the fermentation medium was optimized by the experiments of uniform design.Foundation and application of the strain screening model: the original strain was introduced mutation after continuous mutagenesis with NTG (N-methyl-N'-nitro-N-nitrosoguanidine),ultraviolet and 60Co-γradiation, then the mutants of non feedback restraining of metabolic product were selected with Cephalosporin C gradient dish and at last the strain NUC-118 was obtained as an high-yield strain by pre-screening of agar column and flask selection. The fermentation potency of NUC-118 in 30L fermentor was increased by 25%.Optimization of fermentation medium: Uniform Design is a highly efficient experimental method. After the main components of fermentation medium being optimized with Uniform Design, the CPC fermentation level of NUC-118 was increased by more than 6% in 30L fermentor.Desorption from porous polymeric adsorbents: comparing the gradient and homogeneous desorption, confirm the gradient desorption can improve not only the purity of desorption liquid by 3.23% but also the yield by 2.3% after the adsorption of fermentation filtrate by porous polymeric adsorbents.spray drying of concentrated solution: the spray drying as a more environment-harmonious drying method was discussed in the paper, and the final approach is to replace the current crystallization method consuming menstruum. The result indicated that the salable product of CPC sodium was obtained by spray drying and even the yield was improved. So the spray drying is doable for actual production.
