Porphyrin and metalloporphyrin existed in nature widely and played an important role in life activities. It has extensive application in the biomimetic chemistry, photoelectrochemistry, analytical chemistry and medical science for its rigidity structure and good electronic flowing capability. It has important significance to develop and study novel porphyrin compound with special active functional group.In this paper, the synthesis method for porphyrin and metalloporphyrin were discussed. The application of porphyrin in many fields and development trend were also studied. And the development of probe for bimolecule and drug were investigated at the same time. This paper including two aspects:1. Synthesis of porphyrin compound:Four kinds of novel azo-benzaldehyde were synthesized by coupled reaction and then reacted with pyrrole to produce four new azo-prophyrins. The compound structures were characterized by infrared(IR) spectroscopy, ultraviolet-visible (UV-Vis) spectroscopy, elementary analyse and H nuclear magnetic resonance (1HNMR). The spectra behaviors of compounds were studied especially. And the quantum efficiency of fluorescence has been measured. The dissociation constant and molar conductivity of them were determined. Chemical calculation was combined with experiment, and computer simulation was introduced to get the optimization three-Dimensional structure. Charge numbers of atoms in the compound were calculated and the reaction mechanism was studied also.Four kinds of prophyrin compounds with different substituted group and corresponding complexes with Zn, Mn and Tb were synthesized. Structures of them were characterized by infrared (IR) spectroscopy, ultraviolet-visible (UV-Vis) spectroscopy and elementary analyse also. The spectrum characters of compounds were studied. Some of these compounds were used for the study about interaction between porphyrin and drug as well as biomolecule. 2. Studies on the interaction between prophyrin and drug and biomolecule.The interaction of prophyrin with drug and biomolecule were studied by fluorophotometry and spectrophotometry. Based on this, two novel methods for the determination of drug and biomolecule with high sensitivity using fluorophotometry have been developed. The influences on the interaction were investigated and the optimal conditions were selected. Furthermore, the interaction mechanism between prophyrin and protein was discussed in detail, including binding constant, binding mode, type of fluorescence quenching and the main kind of binding force. CreativenessThe azo-prophyrins and the intermediate product of azo-benzaldehyde synthesized in this article were novel compound that have not been reported before. Properties of these products were completely studied, especially the spectroscopic property, which provided a great deal of useful information to the development and application of porphyrin compounds. The character of porphyrin with substituted group and different metal ions were investigated. The reagents synthesized ourselves were applied in the research of interaction between porphyrin and drug or bimolecular. Two sensitivity methods for determination of drug and biomolecule were established and had been applied to the determination of enoxacin and protein in real samples. The fluorescence quench action of protein by metalloporphyrin were studied. The comparison of fluorescence quench phenomena between BSA and HSA were made.
