This thesis is divided into five chapters. The article has mainly reviewed the electrochemiluminescence (ECL). A novel and sensitive method for the determination of pharmaceuticals has been established using capillary electrophoresis (CE) coupled with ECL detection and the binding of clindamycin with hemoglobin was also monitored using this method.In chapter one, the general introduction of ECL were introduced simply first, then the mechanisms and classifications of ECL were described. In addition to these, the principle of the ECL of tris(2,2'-bipyridyl) ruthenium(Ⅱ) was described and the application of the ECL of tris(2,2'-bipyridyl) ruthenium(Ⅱ) combination technique in pharmaceutical analysis was also given in this chapter. At last, the prospects of ECL were forecasted.In chapter two, based on the enhancing signal of benproperine phosphate on the ECL of tris(2,2'-bipyridyl) ruthenium(Ⅱ), a novel and sensitive method for the determination of benproperine phosphate in oral solution has been established using capillary electrophoresis coupled with ECL detection. Under the optimum conditions, the linear concentration of benproperine phosphate ranged from 1.0×10-7~1.0×10-5 mol/L, and the limit of detection was 1.0×10-8mol/L, the relative standard derivation was 1.1%(n=6, c=1.0×10-6mol/L).In chapter three, a method of capillary electrophoresis with tris(2,2'-bipyridyl) ruthenium(Ⅱ) ECL detection has been applied to the determination of chlorphenamine maleate in vitamin C yinqiao tablet. Under the optimum conditions, chlorphenamine maleate could be separated and detected with in 3 min. The linear concentration of chlophenamine maleate ranged from 5.0×10-7 to 1.0×10-4mol/L. The limit of detection was 5.1×10-8mol/L. This method is simple and quick in operation, sensitive and reliable in determination results and it can be used for the quality control of chlorphenamine maleate in Vitamin C Yinqiao tablet.In chapter four, based on the increasing effect of decloxizine hydrochloride and clorprenaline hydrochloride on the ECL of tris(2,2'-bipyridyl) ruthenium(Ⅱ), a novel and sensitive method for the determination of decloxizine hydrochloride and clorprenaline hydrochloride in Haizhuchuanxiding tablet has been established using capillary electrophoresis coupled with ECL detection. Under the optimum conditions, decloxizine hydrochloride and clorprenaline hydrochloride in Haizhuchuanxiding tablet could be separated and detected with in 3 min. The linear concentration of decloxizine hydrochloride ranged from 5.0×10-7 to 1.0×10-5mol/L and the limit of detection was 1.0×10-7 mol/L. The linear concentration of clorprenaline hydrochloride ranged from 5.0×10-7 to 1.0×10-5mol/L and the limit of detection was 7.0×10-8 mol/L. This method is simple and quick in operation, sensitive and reliable in determination results and it can be used for the quality control of decloxizine hydrochloride and clorprenaline hydrochloride in Haizhuchuanxiding tablet.In chapter five, a novel and sensitive method for the determination of clindamycin in capsules and human urine has been established using capillary electrophoresis coupled with end-column ECL detection, based on the ECL enhancement of tris(2, 2'-bipyridyl) ruthenium (Ⅱ) with the clindamycin analyte. Under the optimum conditions, calibration curve was linear over the range from 5.0×10-7~1.0×10-4 mol/L with a detection limit of 1.4×10-7 mol/L. The binding of clindamycin with hemoglobin was also monitored using this method and the binding constant was estimated to be 3.6×103L/mol.
