In this paper, polysaccharide pullulan was modified with alkylogen. The preparara-tion and property of alkylpullulan, and the release performances of drug-loaded film in vitro were studied.First, the alkylpullulan containing hydrophobic alkyl chains was prepared by way of the active hydrogen of pullulan substituted by alkyl groups. The production had improved its solubility in water. The reaction time, the quantity of alkylogen, the reaction temperature, alkalify time and the amount of sodium hydroxide were discussed. The structure of alkylpullulan was characterized by SEM, FTIR spectrum, elemental analysis and thermogravimetric analysis. As provide a theoretical basis for the application of the field of waterproof material.Secondly , butyl-pullulan film was prepared by the method of solvent evaporation. The best solution medium, the optimal concentration and the best time of breaking the film were received. Meanwhile the butyl-pullulan film was characterized and its properties were determined.Finally, the release behavior of butyl-pullulan loaded aspirin as model drug was studied. The results showed that the release feature of drug-loaded film is corresponded with the principle of slow-releasing drug and the balance cumulative release rate in simulated gastric juice is much lower than in simulated intestinal juice.
