By examining more than decade kinds of resins, 3 kinds of resins were determined, inclding macro-reticular absorption resion DC2 and cation exchanger IA4 which were good at removing pigment and impurity from supernatant fluid as well as anion exchanger IB1 which was used in the neutralization of acidic eluate of IA4. By using the cluster of the three kinds of resins, acarbose was separated and purified effectively. The acarbose product obtained by this techniques of separation and purification reached a purity of 95.1%. Furthermore, the operation conditions of the 3 kinds of resins were also investigated and the process of separation and purification was also optimized.Experiments resits showed that the macro-reticular absorbent DC2 was able to remove the pigment effectively from fermentation broth; whereas the strong acid cation exchanger IA4 had a strong ability to separate the impurity which had a reservation time at 7.5 min and before it detected by HPLC method. By using the two kinds of resins continuously, impurity in the eluate of IA4 was not detected. The elate of IA4 was neutralized by the anion exchanger IB1 , so the pH of the eluate was kept in a appropriate value in which acarbose exist steadily. Then the purified liquid was concentrated by rotary evaporator and the white acerate crystal was obtained by vacuum lyophilization. Finally, the white solid powder of pure acarbose was got by vacuum dryness.Moreover , the conditions of absorption and elution of acarbose by the 3 kinds of resins were also investigated and optimized , and an optimized results was achieved.After continuing separation by the three kinds of resins, the total recovery rate of acarbose in the purified liquid was 88%, and a white solid powder of acarbose ,with a purity of 95.1%, was obtained.Compared with other techniques reported by literature, this techniques was more suitable for industrial production because of its simple in process and low cost. | | Keywords/Search Tags: | acarbose, extraction and separation, resin, static absorb, eluted, purification, concentration and calefaction | PDF Full Text Request | Related items |
