Patrinia villosa Juss which ranks in the genus of Patrinia, has abundant nutrition and great edible and officinal value. On the bases of research papers investigation, the advances in isolation, purification, bioactive function, structure determination, and the polysaccharide research situation and prospect in our country were reviewed. The author conducted a series of study on Patrinia Villosa polysaccharides, which were isolated and purified from Patrinia Villosa, and their phsico-chemical characteristics and chemicalstructure were determined with polysaccharides isolation and purification technique, instrument analysis technique in this paper, and their antioxidation activity were investigated with medical analysis technique. The following results were achieved.(1) Several factors that affecting the extraction process were studied through single factor and orthogonal experiments. It had found that temperature was a significant factor influencing the extraction rate of the water-soluble polysaccharides. The optimal extracting conditions of water soluble polysaccharides from Patrinia Villosa were as follows: the adding ratio of water was l:30,the temperature of extraction was 100℃, the time of extraction was 6 hours. In this condition, the extracting ratio of water soluble polysaccharides from PatriniaVillosa was 14.03%.(2)The residue of water soluble polysaccharides from PatriniaVillosa was extracted by alkali. Several factors that affecting the extraction process were studied through single factor and orthogonal experiments. It had found that the concentration of NaOH was a very significant factor influencing the extraction rate of the alkali-soluble polysaccharides. The optimal extracting conditions of alkali-soluble polysaccharides from PatriniaVillosa were as follows: the adding ratio of alkali was 1:35, the concentration of NaOH was 0.9moL/L, the time of extraction was 5 hour. In this condition, the extracting ratio of alkali-soluble polysaccharides from PatriniaVillosa was 8.53 %.(3) Two Polysaccharides named PVPS-I -H-Fr and PVPS-I -L-Fr were isolated from Patrinia Villosa with water extraction, alcohol sediment, Sevag deprotein, DEAE-52 column and Sephadex G-100 column. Their purity was identified by HPLC with a rate of 0. 227% and 0. 634% (dry weight). Their average molecular weight are 153461.7 and 26085.6 respectively. PVPS-I-L-Fr is composed of five kinds of monosaccharides at least, which contains L-rhamnose , L-arabinose, D-mannose, D-glucose and D-galactose.(4) The in vitro antioxidant capacity of PVPS-I-L-Fr was studied by some assay methods, including the ferric reducing ability (FRAP) , reducing power, scavenging ABTS~+- and hydroxyl radical (·OH). Results showed that the total antioxidant activity of PVPS- I -L-Fr was equal to 4.74% that of Vc; reducing power of PVPS-I -L-Fr increased with the increased concentration of PVPS-I -L-Fr. The ABTS~+- scavenging activity of 0.375 mg/mL PVPS-I -L-Fr was equaled to that of 11.73 mg/kg trolox. The scavenging activity of -OH of PVPS-I-L-Fr at the concentration of 6 mg/mL was 59.77% , which indicated that PVPS-I-L-Fr had an effect of scavenging effect on·OH.The analysis about polysaccharides from Patrinia villosa Juss could provide a theoretical foundation for further development and utilization.
