The proanthocyanidins from seed capsule of Horsebean, a polyphenol compound of many hydroxy, derives from the flavan compound. It is a kind of the living creature flavonoid.The proanthocyanidin has special chemical and pharmacological activity, and can be applied diffusely in many fields.So it is of great significance to research and empolder on it.Research work can be divided into four aspects: (1) Compare the proanthocyanidin extration rate of 50% ethanol, 50% carbinol, and 50% acetone; do research on the effect on the proanthocyanidin extration rate by alcohol concentration,temperature, acidity, marination time, materiel size,solution cubage/weight etc. (2) To separate the proanthocyanidin primarily by acetone and petroleum ether;(3) Do proanthocuanidin static adsorption experiments,static desorption experiments on the big-bore-adsorbing resin D3520,NKA-9,X-5 and AB-8; Do proanthocuanidin dynamic adsorption experiments and dynamic desorption experiments on the big-bore-adsorbing resin X-5 and AB-8;(4) Test the deoxidization capacity of the proanthocyanidins from seed capsule of Horsebean and the inhibitory effects on the oxidation of sesame oil,and the effects of scavenging nitrite ,hydroxyl and ultra-oxygenic anion free radicals, and compare with Vc,BHT.The research results indicate that:(1) With the extraction yield of proanthocyanidins and the selectivity of extraction as optimal condition, the optimal extraction conditions are as followed: solvent 50% ethanol, temperature 55℃, solvent volume to seed capsule weight is 10:1,extracting for 3times,and the time is 2Hr respectively.Extraction yield of proanthocyanidins was about 23.18mg/g..(2) After handled with 1 time volume petroleum aether and 4 times volume acetone, proanthocyanidin's purity reaches 45.8%, and the yield of proanthocyanidins is 97.2% by the process.(3) Compared with 4 kinds of absorbent resins, X-5 resin was chosen as the best resin to separate proanthocyanidins. The experiments showed the optimal conditions of purification were that: the absorptive velocity is 2 bed volumes per hour, desorptive velocity is 1.5 bed volumes per hour; ordinal elution with (5BV), 5%EtOH (2BV), 40%EtOH (4BV).The 40% EtOH eluant was only gathered.In the end, Proanthocyanidins' purity reaches 89.46 % and the yield of proanthocyanidins is 87.03%.(4) Compared with Vc, the proantltocyanidin has stronger capacity in the experiments on deoxidization capacity, scavenging nitrite, scavenging hydroxyl, scavenging H2O2 and scavenging ultra-oxygenic anion free radicals. In the experiment on inhibitory effects on the oxidation of sesame oil, the proantltocyanidin has also stronger capacity than BHT.These experiments proved that the proantltocyanidin has strong anti-oxidation quality, and it is a kind of good and pure natural anti-oxidant.
