Flavonoids were extracted from Helianthus annuus.L by ethanol-extraction, the optional extraction was explored by response surface methodology and conditions were obtained with polyamide resin for purification treatment, the main ingredient of total flavonoids were analysed by HPLC, flavonoids antioxidant properties in vitro and the anti-oxidation of flavonoids in on the soybean oil and anti-bacteria action were studied. The main results were as follows:The ethanol-extraction has the characteristics of high extraction efficiency and low consumption, so it was suitable for flavonoids extracted from Helianthus annuus.L. Based on the single factor experiment.Response Surface Method was used to optimize the experimental conditions. By Response Surface Method the optimum ethanol-extraction conditions were extractions for 2 hours at 80℃ and with 80% ethanol and with the liquid ratio 1:20. Under these optimal condition extract twice, 90.92% of the total extracted flavonoids was obtained.Through the analysis of absorption and desorption capacity and rate, the purific ation technic of three resins were compared. Through the experiments, it could be t hat polyamide resin is suitable for purification of total flavonoids. Its adsorption qu antity of total flavonoids was37.41 mg/g of dry resin and absorption rate and desor ption rate 63.66%, after purification the total content of flavonoids was64.5% in the sample. By HPLC identification, there are 17.2081%quercetin,2.4744%kaempferol,3.2314% and hesperetin,3.1213%isorhamnetin,16.5203% rutin.According to the studying antioxidant activities in vitro, the flavonoids of Helianthus annuus.L was a kind of strong antioxidant. In the same quality concentrations, oxidation resistance is equal to VC. the reduction ability is stronger than VC, the ability of removing DPPH free radical, superoxide anion and hydroxyl radical ability are both weaker than VC.Oven storage strengthen method was applied to research the anti-oxidation on the soybean oil. 0.15% + 0.05% of the total flavonoids of citric acid can effectively will be the shelf life of soybean oil by room temperature 1.178 months to 5.269 months, can be used as a kind of health care function of natural oils used antioxidants.The vitro antibacterial effect of extracts from Helianthus annuus.L was studied. The results show that the flavonoids has stronger effects for E. coli, and have a certain of effects to Staphylococcus aureus, but the inhibitory effects on Bacillus subtilis is not very obvious.
