Sea buckthorn belongs to a kind of shrub or small tree in elaeagnaceae,many nutrient substance and medical composition are barley contained in sea buckthorn fruit and leaves. Because of abundant Light and heat resources in Xin Jiang, the content of every kind of nutrition substance is quite higher. Recently much research has focused on flavonoids in the fruit and marc of Sea buckthorn, The Probe into rich Sea buckthorn leaves resources which has high flavonoids content than fruit is quite little. With Xin jiang wild Sea buckthorn leaves as raw materials, Extraction and purification technology of flavonids was studied in the experiment and its bacteriostasis research.Extraction of total flavones with traditional reflux method. Content determination of total flavonoids in Sea buckthorn leaves by ultraviolet spectrophotometry is 3.11%, which is higher than the sea buckthorn fruit content 0.75%.This study is about extraction technology of flavonoids with microwave radiation, the optimum technological conditions were as follows: alcohol concentration was 60%, extraction power was 60%,extraction time was 3 min ,ratio of liquor and material was 25∶1, pH value was 10 , extracted three times, the extraction yield of flavonoids was 86.6%,The optimal conditions of Ultrasonic extractiorare determined with the single factor and orthogonal design were as follows: alcohol concentration was 60%,temperature was 40℃, the ratio of solid to liquid of 1:25,ultrasonic time was 40 min., extracted three times, the extraction yield of flavonoids was 86.6%.Through the experiment of bacteriostasic activity of total flavones in Sea buckthorn, the results showed that there had strong antimicrobial effects for bacterium. MIC values were: Staphylococcus aureus 0.625%, 2.5% Bacillus subtilis, E-coli 5%.Separation and purification of flavonoids were performed on silica gel, and thin layer chromatography tlc, from the EtOAc fraction of the EtOH we extract 2 compounds, the chemical structures were elucidated by ultraviolet spectrum (UV)and high performance liquid and chromatography (HPLC), they were oquercitrin and rutin.
