Simvastatin is a new cholesterol lowering agent that id partially synthesized from lovastatin. Simvastatin is an inactive lactonization precursor pharmaceutical, it operate by active metabolites or its derivatives via liver hydrolyzation. Simvastatin can be used in curing hypeholesterol. Lots of clinical tests show that simvastatin can remarkably decrease incidence and death rate of coronary heart disease, which is characterized by effective adjusting blood lipid levels, few side effects and good endurance of long term use. Both the clinical tests and the experiment showed that simvastatin is fit for paticients.Simvastatin is one of the most effective LDL-C lowing drugs.In this study, Simvastatin is synthesized with the material of lovastatin, through hydrolysis, selective silylation and acylation, remove the silyl group reactions.And we use trimethylchlorosilane as the hydroxyl protecting agent, 4-dimethylaminopyridine as the catalyst of the reaction, and get a convenient and effective method to produce simvastatin.Meanwhile, we choose the optimical condition of each reaction during the synthesis process.This method is very easy and the cost of manufactures and equipment is very low. Whereas the result showed that both the yield and purity are high. Most of the organic solvent can be recovered in order to be used next time. So this method will have a perfect industrialization foreground.
