Capsaicin compound is the substances responsible for pungency in Capsicum annuum L., which are a series of analogues differing in the structure and character, the most representative is capsaicin. In this paper, on base of the modern technology of extraction and separation, the process of extraction and purification Capsaicin compound from capsicol is studied. The work is described as follows:1. The chromatographic conditions for capsaicin compound were optimized. The optimum detecting condition was determined as follows :MeOH-1%H3PO4 solution (V/V=75:25,) system with 1.0mL·min-1 as the mobile phase, chromatogram column was ODS-C18 (150mm×4.6i.d.,5μm), injection volume was 5uL,detected at 280nm, 40℃. The method is applicable for accurate analysis on capsaicin compound.2. The optimum process of acid-alkali extracting capsaicin compound was studied. The optimum extraction process was selected with investigating pH, the NaOH solution concentration and dosage, extracting time, temperature, and the ratio of stock to liquid. Optimum extraction condition was determined as follows: Capsicol was dissolved in a solution of 10% NaOH by 1g to 3ml on the inventory rating, extracted about 5 hours in 60℃, stewing , the under stratification pH was medicated to 5 by thin hydrochloric acid ,extraction and crystallization, yield 70.64%.3. The process of extracting capsaicin with ethanol was optimized by orthogonal test. Optimum extraction condition was determined as follows: 50 vol. % ethanol as extracting solvent, stock ratio 1:3 (m/V), temperature 60℃, time 3 h.4. Ion-exchange method,acid-alkali extraction and crystallization of organic solvent were used to purify capsaicin from crude material containing 60% capsaicin. Results was showed as follows:(一) Capsaicin compound purity was 94% by acid-alkali extraction which was simple and yield was high. It was better than crystallization with organic solvent.(二) Yield of capsaicin compound was 50.7% , purity was 97.6% by 201×7(717) strong basicity styrene anion exchange resin method.
