Flavones of Hippophae rhamnoides have been studied widely and deeply and 32 flavone compounds have been extracted and identified at abroad. But there were only a few domestic studies of flavones of Hippophae rhamnoides, especially basic research of Hippophae rhamnoides var. sp from Russia. This paper studied extraction and purification flavone compounds of Hippophae rhamnoides var. sp, and identified primarily flavone compounds of Hippophae rhamnoides var. sp by using UV spectrum, TLC and HPLC.1. This paper studied systemically pretreatment technics of Hippophae rhamnoides var. sp, and technics of extracting total flavones, such as water extraction, Soxhlet extraction and ultrasonic extraction. The optimal extraction method ultrasonic was found by using traditional and response-surface analysis method, and optimal extracting parameter was found which is ethanol concentration 79.3%, ultrasonic power 928w, ultrasonic time 27.4rain, solid to liquid ratio 15.3:1, total flavones yields 10.41 mg/g.2. Extracting and purifying total flavones of Hippophae rhamnoides var. sp were studied by using respectively TLC and column chromatography. Optimal developing agent combination of TLC was found out, which is methanol: ethanol: phosphoric acid 79:19:2, and three compoundsâ… ,â…¡,â…¢were found by using column chromatography whose optimal conditions is flavone sample to polyamide ratio 0.2:7, elution speed 2.5mL/min, gradient elution using respectively ethanol concentration 30%, 50%, 90%.3.
