Sea buckthorn is a sort of medcinal plant that has high value. Our country has abundant resource of sea buckthorn. There are large numbers of fruits of sea buckthorn used to squeeze syrup or squeeze oil. The marc that have abundant flavones was regarded as wastes. The marc of sea buckthorn as material, total flavones of Hippophae separating, purifying and its oxidationresistant effects were studied in this paper.On base of studying of other people, the methods of analyzing total flavones of Hippophae by spectrophotometer and HPLC were founded. The effects of different solvents, degreasing on the extraction were investigated. The optimum technological conditions were attained by orthogonal experiment as follows: degreasing with benzinum first, extracted with 65% ethanol, extraction temperature 80℃, the ratio of material to solvent 1: 8 (g/ml), extracting for 3 times(1h for each time). Total flavones was also extracted with the aid of ultrasonic wave, and the results showed that marc treated for 15 minutes by ultrasonic wave before extracting, the amount of the solvent used was greatly decreased and the extracting time was shortened. The purification technology that the flavones were refined by macroreticular resins first and by solvent extracting later was adopted. X-5 macroreticular resin was selected from several macroreticular resins. The experiment results showed X-5 macroreticular resin had big adsorption of the flavones, and the adsorption of the flavones was fast adsorption and followed Langmuir equation. The factors of effecting adsorption and elution were also be researched, such as pretreatment of extracting solution, concentration, pH value, speed of adsorbent solution, effluent speed, concentration of eluting agent in order to find out the optimum conditions. The total flavones of Hippophae were refined by solvent extracting after refined by macroreticular resins in order to attain more pure product. The oxidationresistant experiments were studied by refined flavones, such as reducing power, antioxidation actions to axunge, scavenging action to superoxide anion, scavenging action to hydroxyl free radical. And the results showed total flavones of Hippophae had well oxidationresistant ability.The extraction and purification technology adopted in this paper is simple and operable, and the cost of manufactures and equipments is low, whereas the results showed the yield, purification and activity of total flavones were high. Then the extraction and purification technology of this paper has a wide industrialized foreground.
