| Preparation and functional analysis of cosmetic containing polysaccharides of Eucheuma striatum were discussed in this paper.Eucheuma striatum polysaccharides (ESP) were extracted by hot water and deposited by ethanol. Some of the physicochemical properties of ESP were determined. The compatibility between ESP and materials of cosmetic was tested. As a result, ESP performed stably at the pH value of 4.5 to 8.5, and was compatible with anionic and nonionic surfactants. ESP weakened the thickening effect of some thickeners such as acrylic acid polymer and acrylamide polymer.Oil-in-water formulation containing ESP was designed and optimized by the orthogonal tests. A steady and refined formulation was prepared according to stability tests and organoleptic tests by volunteer. Ingredients of the formulation are as follows: cetearyl alcohol, mineral oil, caprylic/capric triglyceride, squalane, dimethicone, cyclopentasiloxane/cyclomethicone, polyoxyethylene sorbitan monostearate, sorbitan monostearate, glycerin, 1,2-propylene glycol, acrylic acid polymer, sodium hydroxide, disodium EDTA, ESP, diazolidinyl urea/methylparaben/iodopropynyl butylcarbamate, d&c green no.5, fragrance, deionized water.The moisturizing efficiency of ESP and ESP cream was evaluated by weight measuring. As a re.sult, the moisture absorption rate of ESP was higher than glycerin and sodium lactate under the relative humidity of 80% and 40% in 24 hours, and the difference of moisture absorption rate between ESP and hyaluronic acid was not significant. The moisture retention rate of 0.5% ESP cream was higher than 5% glycerin cream and sodium lactate cream, but lower than 0.5% hyaluronic acid cream. The result indicated that the moisturizing efficiency of ESP was relative steady and long-lasting. The anti-herpes simplex virus 1 (HSV-1) activities and anti-coxsackie B3 (Cox B3) virus activities of ESP and ESP cream were tested by cytopathic effect reduction assay and MTT assay. As a result, ESP exhibited potent antiviral activities against HSV-1 and Cox B3 virus, with IC50 values of 15.62μg/mL and 0.98μg/mL.Particularly, the anti-Cox B3 virus activity of ESP was better than ribavirin which ran in parallel as the positive control. Preparation with over 1% content of ESP also exhibited antiviral activities against HSV-1 and Cox B3 virus, and showed low cytotoxicities on the cells with CC50 value of over 2000μg/mL. But the influence of cosmetic ingredients on antiviral activities couldn't be ruled out.The effect of ESP and ESP cream on the healing of dermal wound was observed by the model of skin-wounded rats. As a result, the difference among 1% ESP solution, solvent control, 1% ESP cream and cream-matrix control on the healing of dermal wound was not significant in morphology. The result did not indicate that ESP and ESP cream had advanced the healing of dermal wound in the range of experimental concentrations, but they did not show toxicities on the wounded skin. |
