ABSTRACT: The micro-transformation technology showed a promising prospect in industrialization. At present,our country still was at the start stage in this aspect. This topic goal is to product biodiesel oil raw material with the corn stalk which abandons through microorganism transformation technology, and has carried on some beneficial explorations to its craft and the working condition, Therefore has the important theory and the practical significance.In the paper, Trichosporon rmentans were selected from nine high oleaginous fungium and microbes strains. The mycelium growed fast and had big fat particles. There was a biomass of 24.62 grams (dry) per liter fermentation liquid, fat-producing ratio was 54.25%,and fat-transforming ratio was 9.7%.By the flask experiments of several single conditions which affect TR growth and fat synthesis,the optimal fermentation conditions were decided as follows:the age of fungus was 22hr,the concentration of initial glucose was 12%,C/N was 78:1,pH5.8,the amount of inoculation was 10%,the temperature was 27℃,the period of culture was 4d,the added amount of ethanol was 0.2%.When TR was cultured under this circumstance,the content of fatty acids could reach 60.2%, the biomass was 31.26 grams (dry) per liter fermentation liquid,and fat-transforming ratio could reach 15.68%.Through analyzing the fermentation conditions of the fat production with the hydrolysis of the corn stalk pretreated by dilute sulfuric acid,the optimal fermentation conditions of the fat production by 2.1368 were obtained. The following was the optimal condition: fermentation temperature was 27℃,fermentation period was 4d, pH5.5,the amount of inoculation was 10%, rotational speed was 150r/min.The content of fatty acids could reach 31.45%, the biomass was 24.52 grams (dry) per liter fermentation liquid,and fat-transforming ratio could reach 7.81%.Eight fatty acids including palmic acid,oleic acid,linoleic acid, etc were identified as the main components. These were basically satisfied the high-purity biodiesel raw material request.
