Study On The Screening And Fermentation Technology Of Several High-yield Lipid Fungi

As a kind of new oil resource with good prospectand , microbial oils has being a research focus. In this study,a preliminary selection on lipid producing capability of twelve fungi was made. According to the lipids contents and extraction effect of fatty acid composition, a simple and efficient method was determined, on this basis of which, we studied the fermentation technology of lipid with two fungi which had been selected. The results were as follows:The lipids contents of M.isabellina and M.ramanniana were 24.25% and 21.66% separately,the lipids content of M.racemosus was 18.74%. There were L.starkeyi and R.glutinis which lipids contents were 19.07% and 18.01% ,and the other species were all lower than 11%. Gas chromatography analysis shows that the lipid acids of four of them are mainly C16~C18 which have the maximum content of oleic acid and C16~C18 contents are over 90%. Using M.isabellina and L.starkeyi as the test strains, the extraction effect of lipid by thermal-acid method, soxhlet extraction method and organic solvent method were analyzed comparatively. The results show that thermal-acid method was a simple and efficient method which have excellent effect on extraction of C16~C18. Organic solvent extraction was less efficient. On the basis of the single factor experiment and orthogonal design, the Optimization of fermentation of M.isabellina and L.starkeyi were studied. Under the optimum culture conditions, the biomass yields were 20.25g/L and 16.35g/L separately,the lipid yields were 9.82g/L and 4.94g/L separately,and the lipid contents were 48.49%and 30.21% separately.
