Ganciclovir was the first drug which was used to heal the CMV infection efficiently. It has many advantages, such as reliable curative effect, low virulence and little bad-reactivity etc. It is always used to cure chicken pox and zonal bleb in skin treatment. The main drawback of traditional preparations is their poor bioavailability. Liposome preparation can inosculate with biology membrane easily, promote the quantity of absorbed medication, reduce virulence of medication and prolong the residence time in organism. Therefore, the effect of ganciclovir is promoted on curing skin and eye disease after liposome preparation.1. Set up the Analytical Method of Medication Content in Ganciclovir Liposome.Experimental objective: Determine the dissociative medication concentration and entrapment efficiency of ganciclovir liposome.Experimental method: Set up the ultraviolet spectrophotometry(UV) analytical method of ganciclovir. Entrapment efficiency of ganciclovir liposome was evaluated by exclusion chromatography with Sephadex G-50 and UV. Experimental results: With wavelength at 225nm, it took on a good linear relation between ganciclovir with concentration ranging from 0.2ug ? mL-1 to 20.0μg·mL-1 and its absorbance, the regression equation wasA=0.0705c+0.0071, correlation coefficient r=0.9995. The recovery of separation method were 102.80% and 93.55%.Experimental conclution: Exclusion chromatography of separation method with Sephadex G-50 and UV is desirable.2. Studies on Optimization of Ganciclovir Liposome's Preparation and FormulationExperimental objective: To optimize the preparation and formulation of ganciclovir liposome.Experimental method: Orthogonal design was adopted to screen the preparation (reverse evaporating method) and formulation of ganciclovir liposome based on the monofactor experiments which showed a score according to the entrapment efficiency and leaking percentage.Experimental results: The optimized conditions are rug:lipid=l:12(w:w), cholesterol:lecithin=l:5(w:w), tween-80:lipid=l:15 (w:w), chloroform:warter= l:2.5(v:v). The entrapment efficiency and leaking percentage of liposome, which was prepared on the optimized conditions, are 43.24% and 1.02%.Experimental conclution: Entrapment efficiency and leaking percentage of ganciclovir liposome on the optimized conditions are desirable.3. Preparation of Poly (vinyl alcohol) Coated Ganciclovir Liposome Experimental objective: Improve the stability and entrapment efficiency of ganciclovir liposome.Experimental method: PVA coated ganciclovir liposome was prepared by reverse evaporating—mixing—placed method. The infection of different PVA concentration was showed by entrapment efficiency. Better stability showed leakage efficiency.Experimental results: The entrapment efficiency was improved to 69.63%. The leakage efficiency of PVA coated ganciclovir liposome was reduced to 2.68%.Experimental conclution: The entrapment efficiency and stability of PVA coated ganciclovir liposome was improved clearly. The result is desirable.4. Studies on Character and Stability of Ganciclovir LiposomeExperimental objective: To study the character and stability of ganciclovir liposome.Experimental method: The appearance of liposome was observed by TEM and its particle size was determined by particle size analyzer. The stability of liposome was tested by leakage efficiency and centrifugal acceleration experiment after liposome was placed for a period.Experimental results: The particle size of the ganciclovir liposome was 332.3 nm with the polydispersity index of 0.143. The liposome was stable during the experimental period.Experimental conclution: The particle size distribution of the ganciclovir liposome is symmetrical. The appearance of ganciclovir liposome is regular and the liposome is stable.
