Ginseng aqueous extract liposome was studied in this article. After measuring the total saponins of Panax ginseng in ginseng aqueous extract and the entrapment efficiency, choosing the methods for preparing liposome, one-factor and orthogonal test were used together to get optimum preparation conditions. Then the application performance of ginseng aqueous extract liposome in cosmestics was studied.The content of the total saponins of Panax ginseng in ginseng aqueous extract was measured by the standard addition method . The standard addition method is superior to all other methods for its good precision, high accuracy, rapidity and convenience. The liposome was prepared by film dispersion and freeze-thawing techonology. The free total saponins of Panax ginseng was separated from liposome by dialysis in phosphate buffer solution for 24 hours to calculate entrapment efficiency.The optimum preparation conditions determined by one-factor and orthogonal test were as follow: the ratio of cholesterol and lecithin was 1:6(w:w), the ratio of SPAN80 and TWEEN80 was 1:1 with the total quality of 4.0mg and the ginseng aqueous extract was 0.35mL. The content of phosphate buffer solution was 1/60 mol/L and its pH=6.5. The content of lecithin is 1% in PBS. In this codition, the entrapment efficiency of ginseng aqueous extract liposome was 37.26%.The PVA-coated ginseng aqueous extract liposome was prepared by simply mixing the liposomal suspension with the PVA solution. When PVA concentration was 2g/L, the entrapment efficiency was 46.83% maximally. It is illustrated that PVA could increase the entrapment efficiency of ginseng aqueous extract liposome. The structure determined by electron microscope confirmed the liposome was pellet. By comparison with non-coated liposome whose zeta potential and diameter was -0.9mv and 101nm, the PVA-coated one with -3.9mv and 124nm had larger zeta potential and particle.The physical and chemical stability of liposome were investigated by measuring the changes of zeta potential, diameter, leakage rate and entrapment efficiency. With the time going up, the partical would assemble, the leakage would increase. However, these changes were slower by coating PVA. The vivo experiment proved that they had a good slow-release capability. The best effect of antioxidants on proportion between soybean isoflavone, V_e and BHT is 1.2%:3.1 %: 1.2% on quantity to soybean lecithin’s net weight, by researching their synergistic effect of oxidation resistence.
