Hypericum japonicum Thumb.was the entire dry plants of Garcinia,also be called as Japanese St. Johnswort. The pharmacological activity of Hypericum japonicum Thumb, are widely proved in antibacterial; protection of liver and anticancer.For the sake of better utilization of Hypericum japonicum Thumb. in medicinal area.Study had been carried from the foundation of quality standard for medical material, extraction and purification of total flavonoids, foundation of quality standard for intermediate and preparation of solid dispersion. The aim is to develop a new drug, which has the character of definite effective fraction, enrichment of the active components, well-controlled in quality and used convenient.1,The foundation of quality standard for medical materialQuercetrin was the main constituent in total flavonoids, which was used as the reference substance. Total flavonoe were determined by ultraviolet spectrum(UV) and quercetrin was determined by high-performance liquid chromatography (HPLC)2,The extraction of total flavonoidsThe yield of total flavonoids was used to evaluate the extraction technology through factor investigation and orthogonal experiment design.3,The purification of total flavonoidsUsing the yield and content of total flavonoids as the evaluation standard, suitable macroporous resin was chosen to purify total flavonoids. Other technology parameters were optimized by orthogonal experiment design.4,The foundation of quality standard for intermediateThe contents of total flavonoids and quercetrin were chosen to estimate the quality control of the intermediate, both of the content limitation were established at the same time.5,The preparation of solid dispersionSolid dispersion technology was used to elevate the dissolution of total flavonoids, so as to increase the biological availability of total flavonoids. Using the dissolution of total flavonoids as evaluation index to investigate the type of carrier, the dosage of carrier and the volume of dissolvent. X-ray diffraction and Infrared were used to evaluate the quality of solid dispersion. The hygroscopicity and the flowability of solid dispersion also been studied so as to make base for praeparatum. The result of proximate dissolubility experiment showed that the dissolution of total flavonoids had been increased to 17 times after the preparation of solid dispersion.
