Atractylodes macrocephala koidz (Baizhu in Chinese) is a kind of traditional Chinese medicine. Its rhizome can be used as a medicine with many functions, such as diuresis; diminish inflammatory and fight against senium. Furthermore, recent pharmacological studies also showed that it has anticancer function.Chapter 1 of the dissertation reviewed the studies of Baizhu at home and abroad based on the published papers and articles, including the pharmacological effects as a traditional Chinese medicine, main bioactive components, extraction methods of Baizhu volatile oil and Baizhu polysaccharides, preservation of Baizhu volatile oil, column chromatography separation processes of Baizhu, and proposed the research contents of the dissertation.Chapter 2 studies on the extraction methods of Baizhu volatile oil. The reflux extraction method was selected to extract volatile oil from Baizhu in this dissertation, comparing with ultrasonic extraction method and cold water extraction method. Then the main factors influencing the reflux extraction method was optimized by orthogonal tests, such us extraction solvent, extraction temperature, granularity, the ratio of the extraction solvent to Baizhu powder, extraction time, extraction degree and so on. At last, the optimal extraction technology conditions were found.Chapter 3 studies on the stability of Baizhu volatile oil.β-cyclodextrin (β-CD)-inclusion method was adopted to improve the stability of Baizhu volatile oil. Ultrasonic inclusion method was selected to include Baizhu volatile oil, comparing with water solution inclusion method and rubbing inclusion method. Then the main factors influencing the inclusion process was optimized by response surface methoodology, such us inclusion temperature, inclusion time, ultrasonic wave stength, the ratio ofβ-CD to water, the ratio ofβ-CD to volatile oil and so on. At last, the optimal extraction technology conditions were found.Chapter 4 optimized the main factors influencing the separation process of atractylenolide II, such as solvents, column, silica gels, flow rate of elution solutions, sample loading, and temperature. But even under the optimum conditions, the recovery and purity of atractylenolide II were low. Therefore, the volatile oil was purified by extraction method. And the recovery and purity of atractylenolide II were greatly increased.Chapter 5 summarized the conclusions of this dissertation.
