Jerusalem artichoke ordinarily called "Yang Jiang","Guizi jiang",belong to composite and are the member of helianthus genus,native to North America,and has a long history in China.Jerusalem artichoke has great flexibility and the planting technics is simple,Traditionally people use the fruit of erusalem artichoke as food.Recent years,with the advanced technics it have been processed into Fructo-oligo-saccharide(FOS),Inulin and exceed fructose and have been regarded as nutritional material.The experiment was based on Jerusalem artichoke from Bishan of Chongqing,after analyzing the solicit contributions,Using ferment method can get Endo-inulinase,Which catalytically convert Inulinase to fructooligosaccharides.Through the method of HPLC to determine the product's FOS using sigma's FOS as standard output.After improving the method study the best data for production FOS and putting the product into yoghourt and getting nice purpose.The experiment's result has been showed as follows:1.A series of the condition of HPLC are determined through criterion research.The para- meter as follows:chromatogram column is reverse HypersiL NH2(5μm,250mm×4.6mm),RID-10A SHiMADZu,SPD-10A detector and solvent is acetonitrile:water(72:28),velocity of flow is0.8mL/min, injection volume is 10μL,temperature of the column is 40℃.In this measured condition,the time of apex of 1-kestose,nystose and 1-fructofuranosyl nystose is 15.429 min,20.041 min and 26.128min respectively.The optimal results is 52%(the sum of 1-kestose,nystose and 1-fructofuranosyl nystose),The recoveries were 100.50%,100.11%and 100.31%for capsacin and dihydrocapsacin respectively.Compared with other ways,this method is more rapid, exact,simple and convenient.2.Endo-inulinase catalytically converts Inulinase to fructooligosaccharides and the action mode of Endo-inulinase is switching offβ-2,1-linked polyfructose chains interiorly to get low fructosyl units.Aspergillus Niger is one kind of bacterium which can product Endo-inulinase,The inulinase we have got through ferment is more purify and its I/S>12.Endo-inulinase is extracted by ferment method,the single factor tests,orthogonal experimental designs and variance analyses were applied to optimize the manipulation parameters of ferment method.the result showed:the optimal temperature 30℃,5%substrate concentrates,the best pH value is 5.5,5mL content of zymogen, through the above technical data it can get FOS of 52%,the experiment showed that the method which determined IOS is feasible and is exactitude and repetition.3.Study the extraction process of Fructooligosaccharides,fructooligosaccharides are producted through Endo-inulinase working on Inulin,Fructooligosaccharides of the Jerusalem artichoke is extracted by ferment method,the single factor tests,orthogonal experimental designs and variance analyses were applied to optimize the manipulation parameters of ferment method,the optimal parameter as follows:5%substrate concentrates and the optimal reaction temperature and the proper content of Endo-inulinase is 5mL and the best pH value is 5.5.as following the above method can get the optimal results 52%.From the orthogonal experiment,we get that the first effects of gained ratio is temperatures,second is substrate concentrates,third is the amount of Endo-inulinase,the least one is ferment temperature.Through the HPLC detecting the yield of FOS we know that HPLC is a good which is narrow and repetitious.4.Fructooligosaccharides is a functional oligosaccharide,and has been showed have some benefitial effects such as functional of anticarious,modifying gut microbial populations,stimulating immune function,lowering plasma lipids and helping the absorb of calcium.Acoording to the FOS's technical characteristic during the processing of food making we know during the low temperature and pH the FOS's characteristic could not change more,so accessioning it into yoghourt can make good helpful for people's health and will be a new attempt.After studying on FOS yoghourt we got the proper parameter:2%vaccination,consistent total solid 11%,FOS:Aspartame=1:0.020.
