Surface Enhanced Raman Scattering (SERS) is one of the most regular spectrum technics. Self-assembled monolayers (SAMs) via the artificially design of self-assembled monolayers to attain the aims of controlling the composing, structure and function of the surface. This technic can provide a favorable flat roof for the functional interface at basic research and application of the project.Spectrum electrochemistry is a new technic combining the spectrum and the conventionally electrochemistry. It characterizes and investigates electrochemistry system in situ on the level of molecules. There is a outstanding predominance in investigating the tropism and the bonding of molecules adsorbed on the electrode, and identifying species of molecules participating in the process of electrochemistry. These technics are applied widely in the currently researching field of chemical modified electrode and bioelectricity, and at the same time, these technics are also the primary investigating means for this paper.Actually, there is a less research in the mercaptan ramification monolayers which containing aromatic heterocycle in the molecules than the alkyl mercaptan self-assembled monolayers. Moreover, people give an attention about these compounds constructing monolayers system in the recently researching field of chemical modified electrode and bioelectricity. They have a well foreground in applying. So, we select 4,4′-dithiodipyridine (PySSPy) and azathiopurine(AZA) as our research objects.Concretely to say, this paper deals with the following research works based on the two molecules mentioned above.Experiment indicates that PySSPy SAMs formed on the gold surface can still implement the self-assembled of gold nanoparticles using it′s end. This will come into being a system of self-assembled sandwich configuration with filling, and SERS spectrum of this system with the alteration of voltage and pH value of gold nanoparticles presents a orderliness. This change will help us realize the structure change of PySSPy adsorbed on the substrates with the outside factors. AZA is a ramification of 6-mercapto purine (6-MP). As an immunosuppressant, its effect has exceeded 6-MP. Experiment indicates that the SERS spectrum of AZA adsorbed on roughed gold electrode and silver electrode is different distinctly. We also join SERS with electrochemistry to research the effect of SERS spectrum of the self-assembled system with the change of voltage. Investigation tells us that the adsorption point of AZA on the gold electrode and silver electrode is different. N7 atom is the attachment site at gold surface, and that N3 atom is the attachment site at gold surface. Furthermore, SERS spectrum indicates that the tropism of AZA adsorbed on the two substrates is also different with the change of voltage.This paper will give us a detailed analysis about the SERS spectrum of PySSPy-SAMs and AZA SAMs at different outside conditions, such as different substrates, different voltage and different pH values. At the same time, we will present reasonable academic explains.
