The paper summarized a series of basic research and development on preparation of microbial flocculant. This isolation of flocculant-producing bacterium MF-04,research of substituting culture medium,optimization of culture condition,characteristic of flocculation,application and mechanism of the microbial flocculant were studied and discussed.The main centent of research and the result are as follows:(1)We got microbial flocculant-producing microorganisms from active sludge which was cultivated in specifically culture medium. There were 14 bacteriums, 1 bacterium of which has better flocculation activity, attain 87.1%, denominated MF-04. In order to save cost, we studied the effect of N,P on flocculation activity using sugarcane-juice as substituting culture medium. At the same time, we did research of the compound culture medium.(2)In order to heighten flocculation activity, carbon,nitrogen etc were optimized on the basis of GC culture medium. Elicited sequence of all the effect factor and optimization condition by orthogonal experiments.(3)Elicited optimization numerical value by doing condition validate esperiments in which we studied the impact of MF-04 on pH of kaolin poise,subsiding time,MBF dosage,CaCl2 dosage,whisk speed etc. By the research of growth curve of MF-04, we found that its optimization growth periods was 60h.(4)By distilling and punrifmg the MF-04 producing flocculant, its main component was amylase by infrared spectra and UV, and its content was determined. we found that mechanism was bridge and absorption, -COO- and Ca2+ is single-tooth bridge-linking by zeta potential and infrared spectra.(5)By orthogonal experiments and single-factor, we found that effect of MF-04 producing-flocculants to eliminate mineral wastewater turbidity and COD was prominent. By mixing MF-04 producing flocculant with PAAZ, we found that turbidity was not only faller, but also the flocculant dosage decreased. We found that effect of MF-04 producing-flocculant was better than inorganic polymer flocculants to mineral wastewater by comparing with inorganic polymer flocculant. The effect in decolour of muscovado was prominent by mixing MF-04 producing-flocculant with PDMADDC, and the decolour of muscovado was 38.9%.
