Determination, extraction, decoloration and separtion of shikimic acid were studied in this paper.A New method for determining the concentration of shikimic acid using microwave assisted had been proposed. The experiment conditions had been determined as follows: microwave power 160W, reaction time 2.5min, detection wavelength 382nm.The linear rang of the calibration curve was 0~2.8×10-5 mol/L with r = 0.9992.Extraction experiments were carried out by single and orthogonal experiments. The best condition for the ultrasonic - microwave assisted extraction were determined as follows: the granularity of Illicium verum Hook.f. dusts 20, the ratio of the solid to liquid 1:15, the microwave power 350W, the extracting time 3min. This technology was superior in extraction ration to decoction.The effects of resin dosages and the temperature on decoloration and loss of SA by static experiment were studied. By dynamic adsorption experiments, the best technology conditions were established: temperature 25℃, flow rate 4BV/h, an original pH value. Under the optimal technique, the decolorization ratio was about 75%.Four ion exchange reins were screened by batch static adsorption experiments of SA and static elution experiments of adsorbed SA. Results showed that ion exchange resin D380 presented good adsorption ability. The equilibrium data of adsorption were well fitted to the Langmuir isotherm. The apparent activation energy was Ea = 35.5kJ/mol, the apparent rate constant increases along with the augment of the temperature.The effects of factors such as pH value, concentration, flow rate, the ratio of height to diameter to breakthrough curve were investigated. The conditions were determined as follows: pH value 2.5~3.5, concentration 9.0mg/mL, flow rate 4BV/h, D380 resin dosages 20mL. SA adsorbed on ion exchange resin D380 can be eluted with 1.0mol/L NaOH - 0.1mol/ L NaCl (8:2) solution. Desorption ratio was over 90%. SA was determined by HPLC, And the purity was over 95%.
