Study On Process Of Mevastatin Extraction From Mevastatin Fermentation

Mevastatin is one of the statins, which is usually used as leads of provastatin. Provastatin as a competitive suppressant to 3-hydroxy-3-methyl-reductase inhibitor (HMG-CoA reductase),is very useful to decrease cholesterol. Quality of mevastatin is directly related to transfer ratio of provastatin transition strain and the purity of final product . And mevastatin is the main cost in provastatin manufacture. So a low cost and high quality manufacture process of mevastatin are very important to increase the yield and purity of provastatin.In this article, solvent extraction was used firstly to detect the stability of dried fermentation mycelium and analysis the influencing factors to a rising content of impurity D in the mycelium which was air-dried under different operating conditions. This study also discussed ways to get rid of impurity D, such as opening the lectin ring with mild bases, absorbing mevastatin on big porous resins, recrystallization in different solvents. And the whole extraction process with drying mycelium was validated.Moreover, in this article the extraction of wet fermentation mycelium was studied. The proper extraction solvent was selected by lab experiments, and the best extraction condition was obtained by orthogonal experiments. The lactone was realized by vaccume evaporation the condensed liquor. And the proper condition of close-ring was also obtained. This article also studied the refining process of mevastatin, and the proper solvent system of refining process was selected out.According to the results of lab studies, large scale manufacture of mevastatin was studied. The results expressed that extraction with wet mycelium is proper to mevastatin manufacture, the total yield of manufacture process is 64.1%, and the quality of final products reached standards of overseas users.After all, the results of this study gets to the aims of this project.
