By natural wort flat selection, a high lycopene yield and hereditary character stable Blakeslea trispora strain was obtained.The batch fermentation condition of Blakeslea trispora strain in shake flask was studied. Based on the single factor experiment of fermentation medium, Firstly, among the components investigated, several more important factors were selected by Plackett-Burma. Then, the path of steepest ascent was used to approach the optimal region of lycopene production. At last, the best level of three important factors was determined and the fermentation conditions were studied by response surface analysis.Under the optimum conditions, the yield of lycopene could achieve 1.24g/L by flask-shaking batch fermentation, increasing by 55% than that of initial conditions which was 0.8g/L.. At the same time, from the aspect of comprehensive utilization, amino acid quantitative analysis of Blakeslea trispora was conducted , which provided a scientific basis for exploitation and development of high quality feed.The effect of key enzymes on lycopene production were studied in lycopene metabolic pathway by strain of Blakeslea trispora. Meanwhile, the capabilities of cell growth and production of lycopene were enhanced by addition of activity accelerator of key enzymes. The results showed that the internal and external factors such as reagent A, penicillin, surfactant (span-20), oxygen carriers (n-hexane, H2O2), and other structural analogues of three spore acid (beta-ionone) could effectively promote the production of lycopene by Blakeslea trispora. Among them, the increasing amounts of lycopene were 31%, 32% and 35% respectively by the addition of reagent A(0.05%), beta-ionone(0.1%) and Penicillin(1 mg/L) respectively. But the effect of the oxygen carrier and surfactant is not very obvious.The optimal addition time and concentration of H2O2 and span-20 were 48h ,0.05% and 0h, 0.1% , which were16.7% and 22.7% higher than those without addition respectively.Moreover, the effects of different pretreatment methods of Blakeslea trispora wet cell on the extraction and purification of lycopene were investigated. From the experiment, the pretreatment method of saponification, enzyme and ethanol is better than any other methods. Meanwhile , this method could also decrease the drying time in the process of extraction and purification, which provided basic reference for industrial production of lycopene. As for the lycopene extraction process, results:The optimal extraction conditions were as follows: acetone as solvent, the extraction temperature at 40℃, weight/volume ratio of the sample to solvent 1∶200, and extraction period for 2 hours.
