In this paper, the research focused on probing tritepene saponins, polysaccharides and their active functions.The roots of P. sibiricum Redoute were weighted for 25kg and cutted up. The 70% ethanol was 6 times as much as raw materials decocting 2h. The above experiments all repeated for 3 times. The ethanol extracts were extracted by petroleum ether and butanol . The butanol fraction was isolated by the column chromatography on macroporous resin AB-8, ODS and MCI with ethanol gradient elution and TLC. Two compounds were obtained.Their structures were identified by NMR spectroscopic analysis. Their structures were elucidated as 2β, 3β(OH)2 - (28→1) Glucoside - (6→1) Glucoside - (4→1) Rhamnoside - ursanal (asiaticoside) and 2β, 3β, 6β(OH)3 - (28→1) Glucoside - (6→1) Glucoside - (4→1) Rhamnoside - ursanal (madecassoside). The two compounds were isolated from roots of P. sibiricum Redoute and both were not reported.The crude polysaccharides were extracted 3 hours by hot water, and precipitated 3 times by ethanol, then washed several times with 100% ethanol. The experiment of polysaccharides on the hypoglycemic effects showed that polysaccharides from P. sibiricum Redoute had apparent functions of decreasing blood sugar to alloxan-induced hyperglycemic mice.The polysaccharides were purified by column chromatography on macroporous resin AB-8, and divided into three parts. Then part A was purified by column chromatography on DEAE-cellulose, Sephadex G-100 and HW-40. Moreover, TLC was used for the track examination. One polysaccharide product was obtained. Its structure was determined by GC and NMR spectroscopic analysis. Its main chain was linked by ketoses withβ-1,2 bond.
