In this paper, bio-fermentation method and membrane separation technology had been used to product glycyrrhetinic acid (GA). GA detection methods had been established, systematical researches had been done on the microbial flask culture, seed cans fermentation process and fermentor transformation process. Comprehensive discussion had been done about the membranes selection, membrane impurity and concentration process routes, GA refining process.1. The HPLC synchronous detected conditions of Glycyrrhizic acid (CA) and GA were decided. The correlative coefficient R2=0.999983,standard deviation of the repetitive experiments was 0.00088, relative standard deviation was 0.00061%. The method was exact and reliable.2. CA transformation rates had been used as indicator, determined every process parameters about microbes flask culture, seed cans fermentation, 20L and 200L fermentor transformation process. The biomass received from seed cans fermention was 33% higher than flask culture. The CA transformation rates of flask culture, 20L and 200L fermentation was 78.62%, 77.34% and 74.72% respectively.3. Studied on impurification and concentration of GA fermentation broth with membranes. The effections on fermention broth with different membranes had been discussed. The results showed that UFâ… can get high proteins and solids removal rates used in fermention broth and NFâ…¡c an get better-performing on the concentrated effection of GA. Finally, we can conclude that UFâ… -NFâ…¡two-orders-membranes peocess was the best method used in impurifing and concentrating GA fermention broth.4. Studied on refinement of GA. The pretreatment process of refining GA and every process paramaters were indentified. The styrene-D101 nonpolar macroporous resin was choosen to purify GA, and the best-eluting programme was found. The purity of GA can get 97.50% by pretreatment process, after adsorption and elution by macroporous resin, the purity of GA can get 98.97%.This research was used bio-fermentation method to product GA, the GA fermentation broth was impurified and concentrated by membrane separation technology, the crude GA product then refined by maroporous resin, through all these processes, we can get high up to 98.97% purity of GA products. This process route had the advantages of low cost, high yield and small pollution, and resolved the problems such as highly raw material consumption, difficult manipulation conditions of acid solution, long process route, low product purity, serious environmental pollution caused by acid solution method in our country and all over the world nowadays, and achieved the purpose of clean production.
