Total sugar, also called solid glucose or the industry solid sugar, is made from starch through liquefaction and saccharification. It is mainly composed of alpha-glucose and beta-glucose. The raw material producing total sugar mainly is the starch or high grain containing high starch like corn, the rice, the sweet potato and so on. The total sugar powder taste is sweet, the color is pure white and does not get damp, does not absorb moisture characteristics and so on. The sugariness of total sugar powder sweetness only accounts for 70% of sucrose, equal heat with sucrose, easy absorption by human body, beneficial health. The production the total sugar powder is short, low cost and widely applied in the production of candy, the canned food, the drink, the cakes and pastries, and so on, even can completely or the partly be a substitution of sucrose and the glucose.Corn is a very important kind of resources in industry, which was often used in food manufacture, animal feeds, chemical industry, seed et al. Nowadays over hundreds of products are divided into different series that are made from corn. As for the moiety of the capsomere and strain, corn has a widely usage. Products from retreating also frequently used in further products in food manufacture, chemical industry, fermentation industry, medicine, spinning and weaving industry or making paper et al, As Jilin province is a main place of corn planting and cropping in china, corn's further exploitation will contribute the development of food industry, such as the correlative industry of beverage, cakes and pastry, milk, meat, condiment and nutritional foodstuff, functional food, enzyme preparation, additives and brewing. It even facilitates architecture paint and petrochemical industry, as it does to the industries of paper making, spinning and weaving industry and medicine. Moreover, it push about the development of agriculture, making foodstuff selling more easier, fully use of cropland and generalization the use of modern raise technology. Corn's depth processing will flourish animal feeds industry, prologue industry, poultry raising and fishery.The use of Corn affect all aspects of industry and agriculture, thus the enhancement of the multiple utilization and the further exploitation of corn is meaningful and will elevate the social and economic efficiency.During the whole processes of making total sugar, liquate, saccharification, decolorization and ion exchange are the key links which affect the total sugar quality and the cycle of produce, This study use corn as raw material, aiming at elevating the quality of total sugar, less time consumption, cutting down the cost and make the proceeding more efficiently. The liquefied craft was firstly investigated in this article, and the alpha-amylase was used as the liquefied enzyme, the DE value and the transmittance as the weight target. The effect of the single factors such as the corn density, the amount of thermo-stable alpha-amylase, the liquefied time, the temperature, the pH value and the amount of calcium chloride were studied at details. The optimized parameters were as follows: the corn density 25%, the amount of thermo-stable alpha-amylase 20u/ml, liquefied time 50min, the temperature 100℃, pH value 5.9, the amount of calcium chloride 0.1%.Based on the above research, the DE value and the transmittance were selected as the weighing target, the choice liquefaction time, the temperature, the amount of thermos table alpha-amylase as the research factors, the L9 (34) orthogonal experiment design was carried on. Finally the optimized craft parameters were obtained as follows: The liquefaction temperature 100℃,the amount of thermos-table alpha-amylase 24u/ml,the liquefied time 70min.Through a confirmation experiment, the DE value reached 19.87%,the transmittance 39.46% foreign matter as protein and so on in liquefier are well agglomeration.With regard to the saccharification craft, the DE value is also as the weighing target. The saccharification was carried on through the coordination of the maltogenic amylase and the Prew blue enzyme. Through the single factors experiment, the optimized conditions for the saccharification were that: temperature 59℃, the saccharification time 48 hours, the pH value 4.8, the amount of maltogenic amylase 300u/ml,the amount of Prew blue enzyme 0.1aspu/ml.The response surface design was employed to conduct the optimized the saccharification craft. According to two multi-dimensional regression equation(model):Y=85.87 + 3.05X1 + 5.93X2 + 18.97X3 - 0.1X1X2 - 1.7X1X3 +0.44X2X3-14.25X12-7.32X22-3.86X32,the optimized parameters were obtained as follows: The temperature 59.6℃, time 49.3h, amount of maltogenic amylase 328.6u/ml.Based on the above conditions, the DE value in the saccharification craft reached 98.3%.thereby elevating the efficiency of saccharification, providing a new foundation for producing total sugar.Adopting absorbite as decolorization media, using module of decolorization as measure index, optimize the decolorization parameters, taking the decolorization rate as the weighing target, through the single factor experiment, the optimized conditions were that: the amount of activated charcoal 3%, the decolorization time 20, the decolorization temperature 85℃,pH value 4.6 The response surface design was used to optimize decolorization craft, according to two multi-dimensional regression equation (model):Y=84.86-0.41X1+5.33X2+3.98X3-0.21X1X2+2.65X1X3+1.19X2X3-3.02X12 - 9.45X22 - 7.23X32, the decolorization craft parameters: The amount of activated charcoal 2.59%, the decolorization temperature 79.5℃, decolorization time 29.65min.Based on the above conditions, the decolorization rate reached 85.39%.Deploying ion exchange resin, taking the desalination rate as the weighing target, the flow rate and the ratio of height to diameter were used as the single factors. The regression equation was calculated: Y=73.9-0.02z1-0.48z2+0.0006z1z2+0.0001z12+0.03z22and the craft parameters for the desalination craft were that: flow rate 80, the ratio of height to diameter 7.Based on the above conditions, the desalination rate reached 71.23%.The spray drying technology was used to dry the sugar fluid, and the optimized parameters were obtained as follows: the intake temperature 130℃, the outlet temperature 60℃, the syrup density 55%, the white tiny powder of total powder was obtained. The Uses the high pressure liquid chromatography (HPLC) was used to determine the glucose content of the powdered sugar, the purity of the total powder reached 95.35%.Make linear regression by least square method, obtained the standard curve and regression equation between glucose content and the peak area, with a great degree of precision, reproducibility and recovery rate of application of sample, and the regression equation was showed as follows: Y=6.744621e-0.08)X+8.574846e-0.04.
