Taking scavenging activity of O2- and degree of hydrolysis (DH%) as criterion,the right proteolytic enzyme hydrolysis of buckwheat globulin was studied.Effects of the dosage of protease,the density of substratum,temperature,pH,reaction time on hydrolysis of buckwheat Globulin were studied,and the optimum conditions were determined by orthogonal experiment. The hydrolyzate was separated by Sephadex G-25 gel filtration Chromatography,then studied the antioxidation activity and the molecular weight of each fractions. As the fractionⅢhas better antioxidation activity,then hemolysisi of red blood cells(RBC) and amino acid composition were detected.The result showed that the alkaline protease hydrolyzing buckwheat globulin was better than the papaya protease and the pancreatin,its optimum conditions were:[E] 20000 U/g,temperature 55℃,pH 9.0, substrate concentration 5% and reaction time 2 h.the molecular weight less than 1000 Da has better antioxidation activity,its IC50 of scavenging O2-,OH and DPPH·were 0.75 mg/mL,0.897 mg/mL and 0.38 mg/mL. fractionⅢcould inhibit the hemolysis of RBC significantly,the inhibitory ration was 47.37% with a 1.578mg/mL concentration.Amino acids composition of fractionⅢwas showed to be rich on Glu,Gly,Arg,Lys,Leu and Phe. Glu was the highest amount amino acid in fractionⅢ, and Cys was the lowest.
